Talks between the owners of Makro and unions have failed to persuade the chain from closing three stores.
Makro announced a consultation period last month on the future of stores in Wolverhampton, Swansea and Coventry.
There are 112 staff based in Swansea, 139 at Coventry and 127 at Wolverhampton a total of 378 staff who now face searching for new jobs in a struggling retail economy.
Makro has said it will look at redeploying staff where it can and will offer career support to workers.
A spokeswoman for Makro, which sells homewares and DIY, told diyweek.net: "Consultation talks between (the union) USDAW, representatives from the stores and Makro have taken place and it has been agreed it is no longer viable to continue to operate the Coventry, Wolverhampton and Swansea stores.
"Consultation talks will now focus on supporting the store staff. In collaboration with the union, redeployment opportunities, terms of redundancy and career support will be made available and every effort will be made to find employment for those affected."
